EDX: A Fast & Simple Technique for Pharmaceutical Elemental Analysis

AA or ICP-OES/MS is often viewed as the golden standard to analyze elements during the pharmaceutical manufacturing process, from drug discovery to the QA/QC of final formulations. While extremely accurate, these techniques have the downside of requiring the sample to be in liquid form when analyzing, resulting in a very arduous sample-preparation process and long analysis time. In this seminar, we would introduce Energy-Dispersive X-Ray Fluoroscopy (EDX) as an alternative technique to perform elemental analysis that requires little sample preparation, is easy to use and can provide results quickly; and how it could be used in various areas of the pharmaceutical industry.

 

Validation of Quantitative Method for Determination of Elemental Impurities in Pharmaceutical Products Following USP 232/233 on ICPMS-2030

24 targeted elemental impurities in three drug products were determined following new USP chapters <232>/<233> using Shimadzu ICPMS-2030. The method was evaluated and validated according to the procedures defined by USP <233> and combined with microwave digestion for sample preparation. Good calibration linearity, repeatability, stability and specificity were achieved for all 24 impurities. Low LODs and LOQs were obtained as compared to the target limits from USP <232>. The ICP-MS method applying universal He collision mode provides a high throughput method for routine elemental impurity analysis in pharmaceutical products.

The determination of water content in alcoholic beverages, solvents and various consumer products such as foods, fuels, pharmaceuticals, and petroleum products is one of the most common types of chemical testing. Many techniques such as gravimetric analysis, gas chromatography (GC), Karl Fischer Titration, near infra-red spectroscopy, and others have been used for water quantification with good results. However, the limitations of these approaches can include high limits of detection, large sample sizes required for trace analysis, side reactions, the use of expensive consumables, and the production of chemical waste. Previous work by Prof. Daniel Armstrong and co-workers has described the use of ionic liquid GC capillary columns for the trace analysis of the water content in a wide variety of solvents. These columns were capable of providing a rapid and quantitative determination of water contents using very small sample amounts. This webinar further examines the characteristics of ionic liquid capillary columns for the GC analysis of water.

Monitoring and Quantitation of Nitrosamines in Water using GC-MS via Chemical Ionisation

Formation of nitrosamines, particularly NDMA, is prevalent in water treatment plants using chloramine as a disinfectant. NDMA is classified as a B2 carcinogen based on the induction of tumors at multiple sites in different mammal species exposed to NDMA by various routes. EPA 521 has been provided as the validated extraction and analytical procedure for monitoring of nitrosamines in drinking water. However, this method was developed using large volume injections (20 µL) and ion trap spectrometers in MS/MS chemical ionization (CI) mode (with methanol or acetonitrile). To avoid the complexity of large volume injections and CI, we present the alternative gas chromatography triple quadrupole electron ionisation method. Additionally, calibrants were prepared via an auto-sampler, from 1000 µg/ml commercially available standards.

Nitrosamine compounds are classified as probable human carcinogens by the International Agency for Research on Cancer (IARC). Using MRM/Product Ion Scan Mode in GC-MS/MS to Perform Detection and Differentiation of Synthetic Cannabinoids

Forensic chemists face the challenge of identifying new psychoactive substances (NPS), whereby the general structures of controlled drugs are modified to evade legislative banning. Hence, a plethora of synthetic cathinones and cannabinoids has been available to the public, causing a global social problem. It is therefore crucial that analytical methods are developed for identifying and differentiating new analogues of synthetic drug substances, to prevent potential trading and abuse. Additionally, drug legislation is made more challenging with the use of synthetic cannabinoid containing products, typically marketed as “herbal mixtures”, which consists of analogues of synthetic cannabinoids. In this study, simultaneous multi-reaction monitoring (MRM) and product ion scan (PIS) acquisition methods have been developed for detection and differentiation of isomeric synthetic cannabinoids.

Online Pre-column Derivatization and UHPLC Analysis of Total Amino Acids in Animal Feed

This webinar gives you an introduction on fully automated pre-column derivatization of the amino acids and microwave digestion method which were adopted to further speed up and increase the analysis efficiency. A Shimadzu Nexera LC-40X3 with photodiode array (PDA) and fluorescence (RF) detectors were employed for the analysis. The Nexera LC-40X3 equipped with automated pre-treatment function allows several derivatization reagents to be aspirated sequentially from multiple vials and mixing within the needle and the sample loop. Microwave digestion procedure was employed to carry out both acid and alkaline hydrolysis in less than 30 minutes, which is much faster in comparison to the conventional hydrolysis up to 24 hours. Fluorescent (RF) detection was used for high sensitivity analysis (nM), while UV detection was compared, which shows its applicability for µM level quantitation of amino acids.

Direct Probe Ionization Mass Spectrometry (DPiMS) for High Throughput Analysis

Evolution of mass spectrometry (MS) hardware, especially ionization method, has drastically changed the landscape of MS applications. Direct Probe Ionization Mass Spectrometer (DPiMS) has the potential to pioneer new types of applications with the new ionization interface. The technique successfully incorporated the best of both worlds; high ionization efficiency of ESI and simplicity of MALDI. This was achieved by using an ultrafine needle to sample an extremely small volume of sample on its surface and subsequently applying high voltage to the needle to imitate an ESI probe. DPiMS is characterized by the microscopic sampling volume and high sensitivity for the amount sampled at a low ng/mL concentration range. For this, DPiMS can be a robust solution for the routine analysis of high-complexity matrices, such as plasma or food extract, as contamination of MS hardware can be perfectly mitigated. Moreover, direct sampling from solid samples causing minimal destruction might enable new applications such as real-time analysis of live tissues or cells.

LC-MS/MS methods for vitamin B and D analyses as per AOAC methods in infant and adult formulae

Vitamin B and D are important micronutrients to maintain good health. They are often fortified in various foods such as infant and adult formulae for added health benefits. Over the years, there are many AOAC methods dedicated to the analyses of different vitamin B and vitamin D compounds in infant and adult formulae. In recent years, many AOAC methods use LC-MS/MS to analyse both these vitamins. In this webinar, I will discuss about the different AOAC methods and the sample preparations in the analysis of vitamin B and D. I will also share some of the application development works done closely following AOAC methods using Shimadzu LC-MS/MS.

Nitrosamine compounds are classified as probable human carcinogens by the International Agency for Research on Cancer (IARC). Ever since the first U.S. Food and Drug Administration (FDA) notification in June 2018 about levels of these impurities in pharmaceutical API, they have been a reason of continuous discussion globally for analytical scientist, patients and regulators. After June 2018, the number of recalls and 483’s have been on a rise for not only angiostenin II receptor blockers (-ARB, also called as sartans) but also for other products like Ranitidine, Metformin etc. FDA, European Medicine Agency (EMA) and International Council for Harmonisation (ICH) have constantly set and update the guidelines in measuring impurities in pharmaceutical products. Holistic solution to measurement of nitrosamines need both GC-MS/MS and LC-MS/MS technologies.
